Napier also grabbed 10 fours with a strike rate of 2.62 runs per ball as Essex reached 242-3 and then dismissed Sussex for just 114 to win by 128 runs at Chelmsford.
Despite Napier's exploits New Zealander Brendon McCullum still holds the record for the highest ever Twenty20 score following his 158 not out in the first ever Indian Premier League game earlier this year.
Cameron White had held the previous top score domestically with an unbeaten 141 for Somerset against Worcestershire in 2006.
"It was awesome, fantastic. I just started to see the ball better, middled one or two and then got into a rhythm," said Napier.
