Swede Karlsson, who has managed to finish in the top 10 of all three majors so far this year, continued his good form to lead the way despite hitting a double bogey at the very first hole.
He recovered brilliantly to birdie five of the next seven holes before another at the 11th only to struggle with bogeys at the 14th and 15th holes
Singh, meanwhile, also started poorly but, after a bogey at the first, he then eagled the par-five second before adding three birdies and two more bogeys to his round.
Argentinian Andres Romero was also on two-under when he was forced to finish early because of the weather and will need to complete the final two holes on Friday.
Spaniard Sergio Garcia, who bogeyed the last for a round of 69, is in the group on one under while Phil Mickelson's 70 leaves him a further shot behind.
Open champion Padraig Harrington is on level par after carding a 71 but Lee Westwood struggled in the conditions and could only manage a round of 77 at Oakland Hills.
Elsewhere, England's Justin Rose is two over after a 73, while Ian Poulter and Graeme McDowell are another shot back on 74.
American Kenny Perry was forced to pull out with an eye injury after his round of 79.
