The brothers were broken early in the first set but broke back to force the tie-break and came through when it mattered to take a 1-0 lead.
Both pairings broke serve in the second and so it was down to another breaker, with the Americans running out winners after a marathon shoot-out.
Bob Bryan said: "This is unreal. Words can't describe it. This feels as good as any win we have ever won. We love playing in New York."
While Mike added: "Just a few points (made the difference). I have a lot of respect for Leander, Leander is a legend. And Lukas is an unbelievable player, he is going to win Grand Slams coming up."
